(A) DM199 increases Tregs in pancreatic lymph nodes (PLN). Single cell suspensions of PLN were stained for CD4, CD25, CD127 and Foxp3, and analyzed by FACS. The percentage of CD25+Foxp3+CD127 Tregs within the total CD4+ T cells population was calculated. Data are mean ± SEM (n = 4–10 mice/group). (B) Upper panels: representative images of pancreatic islets stained with anti-Foxp3 (red) and anti-CD4 (green) antibodies, from mice treated for 10 weeks with either vehicle or 100 U/kg/day of DM199. Lower panels: representative images of islets immuno-stained for insulin (brown) and counterstained with H&E. (C) The proportion of CD4+Foxp3+ Tregs in pancreatic islet infiltrates was calculated for each of at least 40 islets per animal.
