Soluble resiquimod does not impact growth of promastigotes or intracellular amastigotes in vitro. (A) Log-phase promastigotes were incubated with increasing concentrations of soluble resiquimod, and metabolic activities (normalized to media alone control) were assessed after 48 h with an MTT assay. Error bars denote the SEM of three combined independent experiments; each experiment was performed in triplicate. (B) RAW264.7 macrophages harboring L. infantum chagasi amastigotes were treated with increasing concentrations of soluble resiquimod, and infected cells were assessed by microscopy after 24 h to quantitate the percentage of cells infected and the average number of parasites per cell.
