DNMT3L facilitates the formation of the protein complex containing DNMT3A, SETDB1, and HDAC1 in E17.5 gonads. Immunoprecipitation was performed with 50 μg of total protein from wild-type or Dnmt3l KO E17.5 gonads. The same amount of protein from control IgG and anti-TRIM28 (A) or anti-HDAC1 (B) immunoprecipitates from wild-type and Dnmt3l KO gonad cell extracts was analyzed through Western blot assays using anti-TRIM28, anti-DNMT3L, anti-DNMT3A, anti-SETDB1, anti-HDAC1, and anti-EZH2 antibodies as indicated.
