Validation of the two-plasmid system. 293T cells were transfected with different proportions of pNCA-S1d (Gag only) and/or pNCA-CAG (Gag-Pol only) plasmid to generate an effective readthrough efficiency from 0 to 12%, and released virus was used subsequently to infect Rat2 cells. TCID₅₀ assays were performed with supernatant serial dilutions as described above.
