Activity of naphthothiazolium salts against Plasmodium berghei in vivo*

Compounds were screened for in vivo activity by using five berghei. C57Bl/6 mice were injected on day 0 with 106 parasitized erythrocytes and then dosed intraperitoneally with 25 mg/kg of the indicated drug at each of the following time points: 2, 24, 48, and 72 hours (n = 6 mice/group). Peripheral blood smears were prepared on day 4 and parasitemia was determined. Mice were monitored daily for 15 days. Inhibition of parasitemia on day 4 = 100 – ([mean parasitemia treated/mean parasitemia control] × 100).16 Percent survival is the percentage of mice that were alive at day 15 post-infection. The five most active compounds are shown.
