2×105 Day 2 sRANKL starved WT and Ads KD (Clone F) cells were seeded in transwells (8 µm) pre-equilibrated in reduced DMEM containing 60 ng/mL sRANKL. The cells were starved for RANKL and then incubated for 20 hours to allow for migration (chemotaxis). After migration, the non-migratory cells on the top of the membrane were removed and migration assessed by enumerating the cells on bottom of the membrane (n = 3). Significantly more Ads KD Clone F cells migrated towards the chemokine sRANKL suggestive of enhanced migration ability in the absence of Ads.
