FIGURE 6. — Inhibition of Tiam1 degradation results in loss of cell viability and increased apoptosis. A and B, HEK293 cells (A) or MDCK cells (B) transfected with HA-tagged wild type Tiam1, HA-tagged Tiam1(S329A/S334A), or HA-tagged Tiam1-C1199 were incubated in low serum for 24 h and then treated with PMA for 6 h. Cell viability was analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The results are the averages of three independent experiments. The error bars represent S.D. C, MDCK cells as in B were incubated in low serum for 24 h and then treated with PMA for the indicated times. The cells were collected, lysed, and analyzed by immunoblotting with antibodies for the indicated proteins. D, as in C except that cells were fixed, stained for cleaved caspase 3, and analyzed by flow cytometry. The results are the averages of three independent experiments. The error bars represent S.D.
