AsPC-1 (A) and MiaPaCa-2 (B) pancreatic cancer cell lines were treated with
AZD1775 (100nM), olaparib (1μM), and radiation (0–6Gy) according
to the schedule illustrated (D). At the end of treatment cells were processed
for clonogenic survival. Data are from a single representative experiment
(A–B) or are the mean radiation enhancement ratio (RER) ± SE for
n = 3 independent experiments (C). Cytotoxicity in the absence of
radiation treatment was calculated by normalizing the plating efficiencies of
drug treated-cells to non-drug treated cells where plating efficiencies for
non-drug treated AsPC-1 and MiaPaCa-2 cells were 0.2 and 0.3, respectively (C).
