A working model of Baf250a in maintaining identity of SAN. The transcriptional hierarchy starts with Baf250a, which activates the expression of Tbx3. Baf250a, Tbx3, and HDAC3 act combinatorially to repress the expression of Nkx2.5 through histone deacetylation in the SAN. Increased expression of Nkx2.5 after deletion of Baf250a in the SAN subsequently activates the expression of Gata4 and Tbx5 in a self-reinforcing manner. Nkx2.5, Tbx5, and Gata4 eventually activate a contractile cardiomyocyte program including enhanced expression of Kcnj2 in the SAN, which negatively affects pacemaking activity of the SAN.
