Mobility shift of the zupT upstream region by Strep tag FurC. The zupT upstream region was amplified as a 563-bp NotI-NcoI fragment. In a reaction volume of 15 μl, 0.2 pmol of this DNA fragment was incubated for 30 min at 30°C without Strep tag FurC (lane 1) or with 1 pmol (lane 2), 4 pmol (lane 3), 8 pmol (lane 4), 12 pmol (lane 5), 16 pmol (lane 6) or 20 pmol Strep-tagged FurC (lane 7). Moreover, the reaction mixture contained no addition (upper), 100 μM Zn²⁺ (middle), or 100 μM EDTA (lower). M, size marker lane. A silver-stained 10% (wt/vol) native polyacrylamide gel was used. Vertical lines were drawn to allow better orientation.
