RBC from mice transplanted with Sec23bgt/gt FLC do not exhibit a band 3 glycosylation defect or a double RBC membrane. (A) RBC ghosts were isolated from Sec23bgt/gt and WT RBC and fractionated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Coomassie blue stain revealed no difference in the appearance of the RBC membrane protein band 3 in Sec23bgt/gt RBC ghosts. Each lane represents a sample from a different individual mouse. (B) Similarly, band 3 protein appeared indistinguishable on Western blotting between Sec23bgt/gt and WT RBC ghosts. (C) Sec23bgt/gt RBC lack the “double-membrane” appearance on transmission electron microscopy characteristic of human CDAII.
