Secondary bone marrow transplantation experiments demonstrate continued equivalence of Sec23bgt/gt and WT FLC at reconstituting erythropoiesis. BM harvested from mice chimeric for GFP Sec23bgt/gt and GFP+ WT hematopoietic cells were transplanted into lethally irradiated secondary recipients (experimental arm). Control mice were transplanted with BM harvested from mice chimeric for GFP and GFP+ WT hematopoietic cells. (A) By peripheral blood FACS, the contribution of Sec23bgt/gt GFP cells to the population of Ter119+ RBC in the experimental arm was equivalent to the contribution of WT GFP cells in control mice over the course of 18 weeks of follow-up. Peripheral blood analysis was performed on 13 to 15 mice per group. Error bars represent standard deviations. (B and C) Bone marrow cells were isolated from both hind limbs of each secondary transplant recipient mouse. The contributions of Sec23bgt/gt GFP Ter119+ erythroid cells at various stages of differentiation (B) and hematopoietic stem cells (C; ckit+ Sca1+ CD48 CD150 Lin) in the experimental mice were equivalent to the contributions of WT GFP cells in the control arm. Each point represents one mouse. Lines represent mean values for each group. P values were calculated with Student's t test.
