Relationship between the OxPhos and respiratory capacities. ADP-and FCCP-induced respirations were successively assessed in Ca2+-free LKB buffer containing 15 mM glucose. Maximal respiration in the presence of ADP alone and ADP+FCCP were taken as OxPhos and respiratory capacities, respectively. A) OxPhos vs. respiration capacity in INS1E and HEK293 cells. Injections-Succ+ADP: 1nM rPFO with 10 mM succinate and 1 mM ADP; FCCP: 2–3 μM FCCP. B) Effects of Pi concentration on the OxPhos and respiratory capacities of HEK293 cells. Injections- PFO: 1 nM rPFO; Succ+ADP: 10 mM succinate and 1 mM ADP; FCCP: 3 μM FCCP. LKB and KH2PO4 differ only in their KH2PO4 content, which were have 0.4 and 10 mM respectively. C) Relative effect of Pi in HEK293 v/slNS1E cells. Mean ± SD from at least 3 well/group are shown. They are representative of 3 independent replicates.
