FBPase, MDH2, Icl1p, Pck1p, GAPDH, and Cpr1p are associated with vesicles in the extracellular fraction. Total extracts were obtained from wild-type cells starved of glucose for 3 days and subjected to ultracentrifugation. The final 200,000× g pellet fraction was aliquoted, and incubated with or without 2% SDS for 30 min. Samples were re-centrifuged at 200,000× g for 2 h following incubation. Western blotting was used to examine the distribution of FBPase, MDH2, Icl1p, Pck1p, GAPDH, and Cpr1p in the 200,000× g supernatant (S) and pellet (P) fractions.
