(A, B). Ionomycin promotes Etn metabolite release from PNT2C2 (A) but not from PC3 (B) prostate cell lines. (C, D). Ionomycin stimulates GPEtn release from the benign PNT2C2 but not from the metastatic PC3 cell line. Cells were labelled with [1-3H]-Etn as described in Methods. After rinsing, cells were incubated at 37 °C in basal medium containing DMSO vehicle (Inline graphic) or 1 (Inline graphic) or 2.5 μM (▪) ionomycin. Aliquots of medium were removed at the time intervals shown for scintillation counting. Results are mean d.p.m./10 μl medium and are +/−s.d. (n=6). In (C and D), Etn metabolites in media from basal- (Inline graphic) and ionomycin- (▪) treated cells were resolved by ion-exchange chromatography (Methods).
