The ponatinib IC50 values were calculated from the data in Figure 2 and FGFR1 protein levels were determined by immunoblotting and densitometry. The indicated cell lines were submitted to 2 color FISH assays for FGFR1 and CEP8 and the ratios are tabulated. The Cancer Cell Line Encyclopedia SNP data was interrogated and the relative FGFR1 gene copy number (GCN) is presented. FGF2 mRNA and protein were measured by quantitative RT-PCR (Materials and Methods) and ELISA (R&D Systems, Minneapolis, MN), respectively, with mean and SEM presented.
