Figure 5. Fucoidan enhances the proteasome-mediated degradation/ubiquitination of TGFRs in NSCLCs.

CL1-5 cells were treated with MG132 (30 min) followed by incubation with fucoidan (100–200 μg/ml, 3 h) and then harvested for immunoblotting with (A) TGFRI and TGFRII antibodies. The detection of ubiquitin (Ub) levels was assessed by the immunoprecipitation of TGFRI and TGFRII (B) in CL1-5 cells. The cells were pre-treated with MG-132 (10 μM) for 30 min, followed by additional incubation with fucoidan (200 μg/ml) for 1 h. The procedure of TGFR immunoprecipitation was previously described in the Materials and Methods section. The results showed the elevation of poly-ubiquitin expression in the presence of fucoidan; the ubiquitin (Ub) and TGFRI/II levels were measured by western blotting analysis. This experiment is representative of three similar experiments.
