Porcine-derived AAV-LacZ vectors were administered IM at a dose of 1 × 10¹¹ gc/mouse and muscle harvested at various time-points. (a) Muscle was harvested at Days 7, 14, 21, and 28 post-injection. Muscle was frozen with O.C.T. and cryosectioned in 10 μm sections, and stained with X-gal and eosin. Blue cells are indicative of AAV-LacZ transduction. AAV1 was used as a control. (b) AAV-LacZ transduced muscle was harvested at 7, 14, 21, and 28 days post-injection and subjected to a β-gal expression assay. Values represent the mean ± SEM. n = 3 mice.
