After T-cell antigen receptor (TCR) engagement, NAD+ promotes naïve CD4+ T cell into Th1, Th2 and Th17 cells. NAD+ promotes the conversion of Th1-IFN-γ-producing cells into regulatory type 1 cells that co-produce IFN-γ and IL-10 with immunosuppressive properties. NAD+ promotes IL-10 and IL-17A cytokines by Th2 cells. NAD+ does not affect Th17 cytokine conditions but drives the switch of iTreg into Th17 TGF-β-producing cells. NAD+ regulates CD4+ T-cell differentiation through Tph1.
