NE-dependent phagocytosis and killing of L. major Δisp2/isp3 by macrophages requires PKR. A, B) RAW264.7 cell lines stably transfected with empty plasmid (RAW-Bla; A) or with the dominant-negative PKR construct (RAW-DN-PKR; B) were infected with L. major for 3 h, washed, and fixed (3 h) or cultured further overnight (24 h). C, D) Macrophages from 129Sv mice (C) or pkr−/− mice (D) were infected for 3 h as just described, washed, and cultured for 48–72 h. Where indicated, the NEi OMeSuc-AAPV-CMK was added at 10 μM. Experiments were performed 3 independent times in triplicates. Graphs are representative of 1 experiment and indicate means ± sd of experiments performed in triplicate.
