The locations of the nucleosome before (oval) and after transcription (dotted oval), and the positions of the sites for the restriction enzymes MseI or ClaI are shown. B. Analysis of DNA fragments after digestion with the restriction enzymes. DNA or nucleosomal templates were transcribed by T7 RNAPs after formation of the EC-5, and digested with restriction enzymes MseI (M) or ClaI (C). DNA was purified and analyzed by denaturing PAGE. C. Quantitation of the data shown in Fig. 2B. Averages from three experiments and standard deviations are shown. Note that MseI site becomes partially protected after transcription, indicating that nucleosome translocation occurred on a considerable fraction of the templates.
