Lack of GLS1 specifically impaired neuronal differentiation. Human NPCs were transfected by control siRNA or GLS1 siRNA and then exposed to the neuron differentiation medium for 6 days. The cells were fixed and stained with MAP-2 to determine the levels of neuronal differentiation. (A) Representative pictures of MAP-2+ (green) and GFAP+ (red) cells were shown. Nuclei were labeled with DAPI (blue). (B, C) The percentage of MAP-2+ cells (B) and GFAP+ cells (C) was determined by counting the number of MAP-2+ or GFAP+ cells over the number of DAPI+ cells in each microscope field. Images were acquired from a Zeiss META 510 confocal microscope. Magnifications: 20× objective lens. Scale bar=50 μm.
