HEK293 cells were transfected with HCV E1 constructs comprising E1 genes cloned from the indicated HCV genotypes and analyzed by an immunofluorescence assay (IFA). The transfected cells prepared 24 h posttransfection were fixed onto slides with acetone and stained with rat monoclonal antibody to HA at 2 μg/ml, H-111 at 5 μg/ml, or R04, a negative control antibody, at 5 μg/ml. Slides were counterstained with a 0.001% solution of Evan's blue counterstain, and bound antibody was detected with fluorescein isothiocyanate (FITC)-conjugated goat anti-human or anti-rat IgG. +++, strong; ++, moderate; −, no reactivity.
