AAV-GMN has broad tissue tropism and retains heparin-binding activity. (a) The indicated cell lines were transduced with AAV-GMN encoding an eGFP reporter at an MOI of 105 and 24–48 hours later, transduction was assessed by eGFP fluorescence. Scale bars are 250 μm (PC-3 scale bar corresponds to all images except fibroblasts).(b) AAV2, AAV-GMN, or AAV5 were incubated with heparin-agarose and unbound flow-through (FT) and wash fractions were retained. Bound virus was eluted with high salt buffer and all fractions were analyzed by slot blot and immunodetection with anti-capsid antibody.
