Response of roGFP2 expressed in chlamydiae upon exposure to redox agents. HeLa cells grown in 4-well chambered coverslips were infected with roGFP2-expressing chlamydiae and exposed to DTT (10 mM) or H2O2 (10 mM) in Ringer’s buffer at different times during the infection. The 402/490-nm ratio of roGFP2 at the basal level (A) and the relative change of the ratios under the different treatment conditions over time were calculated (B and C). The maximal percentages of reduction and oxidation in the presence of DTT or H2O2 at different times were also calculated (D) as described in Materials and Methods. Each experiment was repeated at least three times, and a representative result is shown.
