OA-induced PI hydrolysis is mediated via PLC-ε. Cells transfected with PLC-ε specific siRNA or control siRNA were labeled with myo-[³H]inositol. Cells were treated with OA (10 μM) for 60 s and PI hydrolysis was measured by ion exchange chromatography as increase in water soluble inositol formation. Results are expressed as cpm/mg protein. Values are mean ± s.e.m. of 4 experiments. ^**p < 0.001 vs. Basal ^##p < 0.001 significant inhibition of PI hydrolysis compared to response in cells transfected with control siRNA.
