Stability and reliability assessments of the assay. (A) The assay was screened by three independent experiments in the presence (75 samples) and absence (75 samples) of rhIL-6 (10 μg/mL) at optimized conditions, and a Z′-factor of 0.68 was determined. (B) The competitive binding of ab47215 to membrane IL-6R expressed at the cell surface in the novel assay (IC₅₀=0.38±0.08 μg/mL) and soluble IL-6R in the traditional assay (IC₅₀=0.36±0.14 μg/mL). The 95% confidence intervals for the mean values were approximately 0.24 to 0.56 μg/mL and 0.27 to 0.47 μg/mL, respectively. The data represent the mean±SEM from three separate experiments.
