Figure 3.

Fluorescence imaging of cell surface markers. HBMSCs were immuno-labelled with antibodies against (a) Stro-1, (b) CD146, (c) CD105, (d) Stro-1/CD146, (e) Stro-1/CD105 and (f) CD146/CD105. Single-labelled populations were tagged with both magnetic antibodies for MACS separation and fluorescent antibodies for post-separation visualisation. Dual-labelled cells were fluorescently tagged for FACS separation. Post isolation, cells were seeded onto tissue culture plastic, allowed to adhere, fixed and imaged following DAPI counterstaining. Picture inserts show corresponding bright-field images.

HBMSC: human bone marrow stromal cell; MACS: magnetic-activated cell sorting; FACS: fluorescent-activated cell sorting; DAPI: 4′,6-diamidino-2-phenylindole.
