After specific transfection, GAB1 mRNA levels were significantly reduced (ANOVA, P < .0001). B, Western blot of phosphorylated p44/42 MAPK (ERK 1/2) (phospho-MAPK) mediated activity by insulin in Ishikawa cells transfected with silencing RNA for GAB1. Normalization of protein expression was done with pan-actin. Ishikawa cells without siRNA for GAB1 (Scramble) or without siRNA (Control) showed an increased response of phosphorylated MAPK after incubation with 100 ng/mL of insulin for 10 minutes, compared with cells transfected with siRNA for GAB1 (siRNA).
