hBMSC Production of Candidate Immune Effectors Underlying the Loss of Immunomodulation in Transformed BMSCs

MLCs were established by coculturing PBMCs isolated from two mismatched donors. Cultures of PBMCs from donor A were used as basal syngeneic controls. hBMSC-0H and hBMSC-5H were added to the MLCs, and the production of CXCL10, IL-8, PGE2, IL-10, 6-keto-PGF1α, and TXB2 was determined in supernatants by ELISA after 48 hr culture. Levels of nitrite were determined using Griess reagent to measure indirectly the secretion of NO in culture supernatants. ^∗p value < 0.001 (n = 6 independent experiments; paired t test student).
