Sphere formation assay using unsorted LoVo cells and CD133+ LoVo cells. The sphere formation ability was significantly enhanced in CD133+ LoVo cells when compared with unsorted LoVo cells with statistical significance (unsorted LoVo vs. CD133+ LoVo cells: mean growth, 100.0% vs. 59.97%; difference, 40.03%; 95% CI, 36.75% to 43.31%; P<0.0001). In contrast, a sphere formation activity of CD133– LoVo cells was considerably reduced when compared with unsorted LoVo cells (unsorted LoVo vs. CD133–, mean growth, 100.0% vs. 117.1%; difference, –17.09%; 95% CI, –22.52% to –11.67%; P<0.0001). The sphere formation was remarkably enhanced in CD133+ cells when directly compared with CD133– cells with statistical significance (CD133+ LoVo cells vs. CD133– LoVo cells: mean growth, 117.1% vs. 59.97%; difference, 57.12%; 95% CI, 50.78% to 63.46%; P<0.0001).
