dNMP and rNMP misincorporation during Mn²⁺-dependent synthesis. Polymerase reaction mixtures (10 μl) containing 10 mM Tris-HCl, pH 7.5, 50 nM 5′ ³²P-labeled 13-mer/18-mer primer-template as specified, 1 μM DinB2, 1 mM MnCl₂, and either no nucleotide (–) or 100 μM of the indicated dNTP or rNTP were incubated at 37°C for 10 min. The reaction products were analyzed by urea-PAGE and visualized by autoradiography.
