RT-PCR and immunofluorescence were conducted to detect the expression of SPAG6 respectively. (A) RT-PCR showed that the 190-bp amplicon representing the Spag6 mRNA was detectable in the middle ears of the wild-type mice. No amplicon was detectable in the Spag6-deficient mice. (B) The immunofluorescence staining showed that SPAG6 (red) is located in the cilia of the ciliated columnar epithelium. (C) No staining could be detected in Spag6-deficient mice. DAPI (blue) and phalloidin (green) staining were used to outline the cell nucleus and cytoskeleton respectively. M, mutant; WT, wild-type. Bars: 10 µm in B–C; 7.5 µm in B'.
