KAT6A plays a role in breast cancer stem cell activity. A. SUM-52 control and KAT6A knockdown cells were plated in triplicate in 6-well low-attachment plates for a primary mammosphere assay. After one week, mammospheres were transferred to standard 6-well plates overnight, stained with 0.2% crystal violet and counted. B. Cells from the primary mammosphere assay were harvested, trypsinized, and re-seeded in a secondary mammosphere assay at 3,000 cells/well. After one week, spheres were processed as above for staining.
