CX3C chemokine fractalkine (CX3CL1) prevents monocyte apoptosis in response to serum-starvation. A, Primary human monocytes were incubated for 4 hours in serum-free medium (SFM; left), in SFM+10% fetal calf serum (FCS; middle), or SFM+100 nmol/L CX3CL1 then stained with annexin V fluorescein isothiocyanate (FITC) and 7-aminoactinomycin D (AAD), and analyzed by flow cytometry. Dead cells were defined as those in the upper right quadrant. B, Monocytes were treated as in A with the indicated doses of chemokine-domain (CKD) CX3CL1 or full-length (FL) CX3CL1, including the mucin stalk. C, Monocytes were treated as in A with the indicated chemokines at a dose of 100 nmol/L. D, Monocytes were pretreated ± AZ12201182 (AZ) and then treated as in A.
