Lmx1b.1 and foxc1a potently induces endogenous nphs2 expression in zebrafish. Transient overexpression of lmx1b.1 and foxc1a was made by the Tol2 transposon-mediated transgenesis (see Concise Methods). The Tol2-based plasmids coexpressing lmx1b.1 and foxc1a at the same time, and expressing the two genes individually, under the control of the universal EF1α promoter, were injected in Tg(podocin:GFP) embryos. An empty plasmid was injected as a control. Gene expression (ΔCt) was evaluated by quantitative PCR and presented as fold changes, defined as mean 2ΔCt in tested divided by that in control.
