MAb 9H10 binds to the stalk domain of H3 HA. MDCK cells were infected with the indicated influenza viruses at an MOI of 3 to 5 for 12 to 16 h without trypsin. The cells were fixed with 0.5% paraformaldehyde and blocked with 5% nonfat (NF) milk. MAb 9H10 was used at a concentration of 5 μg/ml, and a goat anti-mouse antibody conjugated to Alexa Fluor 488 was used as a secondary antibody. Recombinant chimeric viruses cH5/3 and cH7/3 express HAs that have the globular head domain of H5 (VN/04) and H7 (SH/13), respectively, with the stalk domain of an H3 (Perth/09) HA. An MAb directed against the M2 protein (Control) served as a positive infection control.
