siRNA knockdown of endogenous human AP3B1 decreases Nipah VLP production. 293T cells were transfected with a plasmid encoding Nipah virus M protein together with 100 nM siRNA as indicated. (A and B) Endogenous AP3B1 was detected by immunoblotting using AP3B1-specific antibody, and protein bands were quantified using a laser scanner. Results from three independent experiments were plotted, with standard deviations indicated by error bars. (C and D) Nipah VLPs were purified and detected as described in the legend to Fig. 4. Results from three independent experiments were plotted, with standard deviations indicated by error bars. Differences from the values obtained in the presence of the control siRNA were assessed for statistical significance by using a two-tailed Student t test, and P values of <0.01 are denoted by asterisks.
