Visualization of HSV- and PRV-associated particles prepared from the cytoplasm of Vps4A- and Vps4A-EQ-expressing cells. Cells were preinduced (Pre Tet+) or not preinduced (Pre Tet-) with tetracycline for 16 h and then infected with HSV (A, B, E, F) or PRV (C, D, G, H) in the presence of tetracycline to express Vps4A or Vps4A-EQ as indicated. After 16 h, PNS samples were prepared and mRFP1-decorated capsids imaged by fluorescence microscopy in the red channel. Representative fields are shown. Scale bars represent 10 μm. (I, J) Quantitation of particle numbers from fields similar to those shown in panels A to H. For both HSV and PRV, particle fields from five independent imaging chambers were counted for each cell type and condition.
