CD4⁺FoxP3⁻ T cells were FACS isolated from the spleen of FoxP3 GFP knock-in mice and induced to differentiate under T_reg polarizing conditions for 3 days as described in Fig 1. The CD4⁺ FoxP3⁺ T_reg generated in vitro were purified and mixed with CD4⁺CD25⁻ responders (T_resp) labeled with cell trace violet and stimulated with anti-CD3 plus mitomycin C treated APCs.
