Propentofylline and AV1013 inhibited upregulation of TNFα mRNA in response to LPS and M3G in adult rat CNS endothelial cells in vitro. (A) Cells treated with vehicle for 1 hr, followed by 10 ng/ml LPS for 4 hr, exhibit significant upregulation of TNF-α mRNA. This upregulation is completely blocked by a 1 hr pre-treatment with AV1013 or propentofylline. (B) Cells treated with vehicle for 1 hr, followed by 0.1 μM M3G for 4 hr, exhibit significant upregulation of TNF-α mRNA. This upregulation is completely blocked by a 1 hr pre-treatment with AV1013 or propentofylline.
