Human (A) CYP3A4 and (B) MDR1 mRNA expression levels were determined in LS180-hPXR-CYP3A4-luc cells transiently transfected with hPXR siRNA (siPXR) or non-target control siRNA (NTsiRNA) before 48-h treatments with the indicated compounds. (*p < 0.05; in the t-test comparisons were made between siPXR-treated and NTsiRNA-treated samples for each treatment group). (C) Protein expression profile of hPXR from one representative sample in LS180-hPXR-CYP3A4-luc cells upon siRNA knockdown of hPXR expression. Anti-PXR and anti-β-actin were used to detect PXR and β-actin, respectively.
