A: Effect of CH25H on HCV replication. JFH HCV Replicon cells were transfected with HA-CH25H expression plasmid and immunofluorescence staining performed 48 hours later using a rabbit polyclonal anti-HA antibody and a mouse monoclonal anti-NS3 antibody, followed by an FITC-conjugated goat anti-rabbit and a TRITC-conjugated goat anti-mouse secondary antibodies. Red fluorescence shows NS3 staining, while green fluorescence detects CH25H. B: Effect of CH25H on HCV RNA expression. The CH25H expression plasmid or vector control plasmid was transfected into JFH replicon cells. Total RNA was extracted 48 hours after transfection and quantified by real-time RT-PCR. C: Effect of 25HC on HCV replication. Replicon cells were treated with increasing amounts of 25HC (0, 1, 5 uM) for 24 h (C) or 48 h (D). HCV Replicon RNA was quantified by real-time RT-PCR.
