Mesangial cells were pre-treated as in figure 4. (a) Cells treated with HG were treated with different concentrations of AS101 (0.1–2 µg/ml). After 24 h, cell lysates were extracted for western blot analysis detection of p-mTOR; p-(rp)S6, with actin as loading control. Results represent three experiments. (b) Cells treated with HG were treated with different concentrations of AS101 (0.1–5 µg/ml). After 48 h of treatment, cells were stained with PI buffer. Cell size was determined by FACS, expressed in arbitrary units as assessed by Flowjo program under Forward Scattered analysis (FSC). Geometric mean (geo.mean) of cell size in each sample was determined. Results are representative of two experiments.
