(A) Total RNA was extracted from magnetic bead-enriched CD11b+ and CD11c+ BIL populations (purity was > 85% and > 80%, respectively) and evaluated for Ifna6 and Ifnb1 by qRT-PCR. (B) BMDM cells from WT mice were stimulated with gDNA derived from GL261 or NIH3T3 cell lines, or solvent only as mock treatment with (grey bars) or without (black bars) pre-treatment with DNase. After 48 h, total RNA was extracted and evaluated for pan Ifna and pan Ifnb by qRT-PCR. (C) BMDM from WT (black bars) or StingGt/Gt (grey bars) mice were stimulated with GL261-derived gDNA (1 or 5 μg/mL). After 48 h, total RNA was extracted and evaluated for pan Ifna by qRT-PCR. Each experiment was performed at least twice. *p < 0.05 based on Student’s t-test.
