Uridine completely prevents myxothyazol-induced activation of caspases-3/7 in the control cells, but not in the cells with ATF4 knockdown. (a) RKO cells stably expressing either scrambled shRNA or ATF4 shRNA (shRNA1, shRNA2) were treated with 1 μM myxothiazol for 3 h or left untreated. ATF4 mRNA levels were examined by RT-qPCR. (b) RKO cells stably expressing either scrambled shRNA or ATF4 shRNA (shRNA1, shRNA2) were treated with 1 μM myxothiazol or 1 μM myxothiazol+1 mM uridine for 24 h or left untreated (Control). Caspase-3/7 activity was measured as a relative rate of Ac-DEVD-AFC (20 ∝M) hydrolysis expressed as relative fluorescence units per hour (RFU/h). Mean and S.D. are presented of three independent experiments; each value was normalized to the corresponding value for the control (untreated) cells
