PKCη expression has opposing effects on IL-6 and IL-8 transcription in response to oxidative stress and DNA damage. MCF-7 clones expressing sh-PKCη (sh2-2 and sh3-3) or scrambled (sh5-3) control cells were treated with 150 μM H2O2 or 400 μM etoposide for 2 h, followed by the addition of fresh medium for 96 h. Total RNA was extracted and cDNA was generated by reverse transcription as described in Materials and Methods. RNA products were analyzed by real-time PCR according to the manufacturer's instructions. (a) IL-6 mRNA and (b) IL-8 mRNA levels are depicted.
