iASPP regulates the proliferation/differentiation switch in keratinocytes by regulating a miRs/p63 axis. Under normal conditions, p63 regulates iASPP expression, which represses the expression of miR-574-3p and miR-720. miR-574-3p and miR-720 repress p63 expression, providing a feedback loop, which maintains the proliferation/differentiation balance. iASPP knockout keratinocytes demonstrate defective differentiation programmes that are associated with an upregulation in the expression of these miRs and the inhibition of p63¹²
