Bone marrow cells from Stat3f/f, Cre-ER Stat3f/f, Cre-ER Ube2nf/f or Cre-ER Stat3f/f Ube2nf/f mice were cultured in M-CSF-containing media plus 4-hydroxytamoxifen for 4 d to generate Stat3-sufficient (Stat3f/f), Stat3-deficient (Stat3δ/δ), Ube2n-deficient (Ube2nδ/δ) or Stat3; Ube2n deficient (Stat3δ/δUbe2nδ/δ) macrophages, respectively. Cells were cultured in M-CSF- and RANKL-containing medium for 3 d (a) or stimulated with LPS (b), as indicated. Gene expression was analyzed by qPCR. Results represent mean values of 3 independent experiments. Error bars indicate SEM.
