Lineage-depleted (Lin) NR4A1GFP transgenic (Tg) reporter mouse bone marrow (BM) cells were isolated, stained and analyzed by flow cytometry. Non-Tg littermates were used as controls (grey). (A) Relative NR4A1GFP expression levels in hematopoietic subpopulations: Myelo Prog, myeloid-committed progenitors (LinSca-1c-Kit+); LSKs, LinSca-1+c-Kit+ cells; LT-HSCs, long-term hematopoietic stem cells. (B) Proportional contribution of LT-HSC compartment (CD150+CD48) to LSKs with increasing NR4A1GFP expression levels. Data are from four independent experiments, each using 1-2 NR4A1GFP Tg mice and 1-2 non-Tg littermate control mice. P-values were calculated using unpaired, one-tailed student’s t-test.
